Insight Biotechnology is extremely pleased to distribute products and services from Cellider Biotech.  Cellider Biotech started as a spin-off of the Aragon Health Sciences Institute (IACS & ISS-Aragón), thanks initially to collaboration of research groups in regenerative medicine and tissue engineering.  Regenerative medicine has improved treatment and quality of life for patients with many diseases and Cellider hope to further improve treatment options for cartilage and bone related diseases.  Advances in these areas will help scientists design cell therapy treatments for numerous patients suffering a range of pathologies.  Cellider Biotech is also committed to investment in R&D targeted towards design of cell therapy treatments for veterinary applications.

Adipose Stem Cells

Adipose-derived stem cells (ADCSs) are isolated from subcutaneous adipose tissue (a suitable reservoir of stem cells). These cells have the potential to differentiate in vitro into chondrocytes, osteoblasts and adipocytes and have shown their ability to heal wounds and regenerate tissues in clinical trials. ADSCs are isolated from a single animal and cultured until passage 1, and then cryopreserved. Post-thaw viable cells are tested for proliferation and differentiation capacity into adipocytes, osteocytes and chondrocytes.

Dental Pulp Stem Cells (coming soon)

Dental pulp stem cells (DPSCs) can be obtained from primary or adult teeth. These cells, similar to mesenchymal stem cells, are capable to differentiate into chondrocytes, osteoblasts and adipocytes. Due to their origin, they can also differentiate into odontoblasts, cells of the nervous system and muscle cells. DPSCs are isolated from a single animal and cultured until passage 1 and then cryopreserved. After thawing, viable cells are tested for proliferation and differentiation capacity into adipocytes, osteoblasts and chondrocytes.

Chondrocytes are the cellular component of cartilage. Articular cartilage is the soft avascular tissue that covers bone surfaces in a synovial joint. Chondrocytes are isolated from knee or hip joints of a single animal, cultured until passage 1 and then cryopreserved. Post-thaw viable cells are tested for proliferation and their ability to redifferentiate into chondrocytes after being expanded (tested in micropellet culture for glycosaminoglycans release by Alcian Blue staining).

Fibroblasts are stromal cells present in connective tissue with a heterogeneous morphology depending on their activity or location and that show similar surface markers with mesenchymal stem cells (MSCs). These cells are involved in wound healing but in contrast, they do not possess the immunosuppresive effects MSCs have. Fibroblasts are isolated from skin of a single animal, cultured until passage 1 and then cryopreserved. Post-thaw viable cells are tested for proliferation.

Services that Cellider Biotech offers include antibody testing and cell isolation, expansion and characterization. Cellider also offer cytotoxicity assays to evaluate biocompatibility of drugs and chemical compounds. All services can be customized to the customer’s specific needs.

Antibody Testing

Antibodies are routinely used as a tool for specific recognition of antigens in cell and tissue characterization assays. Selection of a suitable antibody is a crucial step for adequate cell characterization and reliable immunological analysis, among many others uses. We provide to antibody-manufacturers laboratories and companies with a customized service for antibody testing, offering the possibility of testing a batch of antibodies in different types of cells. This test will evaluate the ability of antibodies to recognize specific antigens and will detect, if it exists, cross-reactivity between different species, providing customers with all the information regarding the antibody.

Antibody testing service includes among many others the following options:

1. Technique: antibodies may have different efficiency depending on the type of analysis in which they are used. For that reason, we test antibodies for several protocols such as flow cytometry, Western blot, ELISA, immunofluorescence, etc.

2. Titration: assessment of the optimal concentration of antibody for every type of analysis (measured by volume and cell number).

3. Cell type: antibodies can be tested on primary cells, stem cells and blood derived cells, always according to the needs of our customer. This will help to determine cell specificity of the antibodies.

4. Species: possibility of antibody testing in several species, including human, mouse, rat, rabbit, equine, canine, bovine, ovine, feline, and swine.

Cytotoxicity Assays

Products developed by pharmaceutical, cosmetics, chemical, and veterinary drugs manufacturer companies require analysis to determine the biocompatibility with the tissue and species in which the product will be applied. The cytotoxicity assay is a rapid and effective method used to scrutinize drug toxicity and/or pharmaceutical compounds on cells. Quantification of preliminary harmful-dose ranges is very useful before further toxicology testing.

We offer cytotoxicity assays as a screening tool for drugs and pharmaceuticals developed with biomedical or veterinary purposes. For that reason, assays can be performed on cells from different tissues (depending on the drug to be tested) and species (human and animals). Analysis of apoptosis and cell viability can be combined with cytotoxicity assays to elucidate potential mechanisms involved in cell death.

In Cellider Biotech, we help our customers in experiment design to estimate compound dose-ranges, adequate type of cells, analysis methods and any other requirement that may appear during the study. Monitoring side by side with our customer from the beginning to the end of the assay will facilitate a successful outcome.

Cell isolation, expansion and characterization

Cell isolation

To optimize basic and applied biomedical research, Cellider Biotech offers the possibility of customized cell isolation from different tissues including bone marrow, adipose tissue, synovium, dental pulp, cartilage, bone, skin among many others. Tissues can be provided directly from researchers or obtained by Cellider professionals.

Cell expansion

After cell isolation, customers could decide if primary cells are sent to their laboratories or if they prefer their cells to be cultured and expanded under specified culture conditions by the Cellider team. Depending on the cell type, cell expansion may take 1-2 weeks for a single passage. After each passage, 1-2 million cells will be frozen and some others kept in culture upon customer request. Cell growth and proliferation will be recorded. All cell lines are tested for mycoplasma, fungi, and bacteria and guaranteed to be free of any pathogen at the moment of supply.

Cell characterization

Cellider Biotech can characterize cells expanded by Cellider or cells expanded in your laboratory using flow cytometry (analysis of cell surface or intracellular markers), microscopy, or by assessing differentiation capacity to several lineages. For mesenchymal stem cells, approximately two months will be necessary for complete characterization. Surface marker analysis includes the quantification of CD34, CD45, CD44, CD73, CD90, CD105 (any other marker will be included upon request) and at least chondrogenic, osteogenic, and adipogenic differentiation capacity will be performed.
