Sample Preparation
Assays have been validated on samples derived from different cell lines, tissues, and species including human, mouse, and rat. It is recommended for the testing services that Marligen prepares the nuclear extracts from your samples. The following are recommended guidelines for preparing your samples prior to shipment:
- Tissues –Tissue samples should be snap frozen, stored at –80°C until shipping on dry ice.
- Suspension cells – One culture volume of ice cold PBS should be added to suspension cells. Centrifuge gently (<1500g) to pellet cells. Resuspend cells in 1ml of cold PBS, Transfer to microfuge tubes and pellet at 3000 rpm in a microcentrifuge (Eppendorf model 5415G or equivalent. Completely removed PBS from cells, snap the pellets and store at –80°C until shipping on dry ice.
- Adherent cells – Prior to freezing and shipping, adherent cells should be processed using Marligen’s Adherent Cell Lysis (Catalog No. 11984-050) reagent using the supplied protocol.
Prior to starting your experiment, it is highly recommended that you contact Marligen’s technical staff.
If you wish to prepare your own samples, it is recommended that extracts are prepared using Marligen’s Nuclear Extraction Kit. To ensure compatibility of the samples with the assays, Marligen’s technical staff should be consulted if you are considering other kits or protocols for preparing samples. We recommend that you provide 20-30 μg of extracts at a concentration of 2-10 μg/μl to ensure adequate sample is available for optimal results with testing in triplicate. Extracts must be frozen at –80ºC immediately after they have been prepared. Because yields of cells and tissues can vary greatly, new customers may want to consider performing a pilot experiment to verify that sufficient material is available for performing the tests. Our technical experts will discuss this with you before you begin.
Shipping
Samples must be frozen immediately at –80ºC and shipped on dry ice. Each tube should be clearly labeled, and the tubes should be placed in a plastic bag and shipped to Marligen along with a description of the samples and the type of service requested is recorded. Please pack your sample on at least 6 KG of dry ice in a Styrofoam cooler with an outer cardboard container and dry ice labels. Please read and follow these instructions carefully. We recommend that you use Federal Express. The enclosed guidelines are for standard overnight deliveries. Please confirm estimated delivery time, packaging and labeling with FedEx before shipping material. Shipments should be sent on Monday, Tuesday or Wednesday only.

We recommend that you send the name of the carrier, the airway bill number and the expected delivery date for your shipment to arrive to Marligen via email to customer.service@marligen.com. We will notify you when the samples have been received and the contents of the shipment have been verified against your packing slip.
Testing Services
Upon receipt of the shipment samples will be processed or stored accordingly. Unless otherwise requested, the multiplexed transcription factor assay will be performed in triplicate for each sample. Results provided include the raw mean fluorescence intensities (MFIs) as an Excel file, graphs and a report summarizing the results. Typically an electronic copy of a report summarizing the results is transmitted within 10 business days after receipt of the samples.
