Santa Cruz: Post Transcriptional Gene Silencers
Santa Cruz Biotechnology now offers siRNA and shRNA (plasmid and lentiviral) gene silencers targeted to all (100%) known human and mouse genes. siRNAs are provided as pools of 3 target specific 19-25 nt siRNAs designed to knock down expression of specific gene of interest. Each vial contains 3 nmol of lyophilized siRNA and is sufficient for 50 - 100 transfections. For independent verification of gene silencing results, we also will provide, upon request, individual siRNA duplex components, each containing 3 nmol of lyophilized siRNA. shRNA Plasmid DNA Gene Silencers are provided as pools of three to five lentiviral vector plasmids each encoding target-specific 19-25 nt (plus hairpin) shRNA (sequences correspond to our well-established existing product line of siRNA gene silencers). shRNA Plasmids are provided as 20 µg transfection-ready plasmid DNA for either transient or stable (puromycin-selection) target specific gene silencing. shRNA Lentiviral Particles are provided as pools of three to five expression constructs each encoding target-specific 19-25 nt (plus hairpin loop) shRNA and are provided as 200 µl transduction-ready viral particles for gene silencing in mammalian cells (human or mouse). Suitable control antibodies and RT-PCR primers are available for monitoring gene expression knockdown.

Origene: HuSH-29: Genome-wide shRNA for Human, Mouse and Rat Genes
HuSH-29 are pre-designed shRNA with genome wide coverage (human, mouse and rat) that delivers guaranteed knockdown. The same superior design is also offered for other species (please enquire).

Allele: RNA Interference
Allele Biotech was one of the very first companies to offer synthetic siRNA and RNA polymerase III promoter systems for expressing shRNA. Allele Biotech has been granted US patents on DNA-based siRNA and shRNA platforms that are popularly used as gene silencing tools. Allele Biotech´s SilenCircle platform design allows high level expression of siRNA or shRNA. RNAi transcripts expressed by this method have sequence identities precisely as desired, i.e. without extra bases introduced by cloning sites, thereby reducing non-specific effects. The LineSilence platform depends on linear DNA cassettes for expressing siRNA, eliminating cloning steps. The AlleleHPLC line of RNAi products offers the best value in synthetic siRNA currently on the market, with guaranteed amount of annealed siRNA, every single siRNA HPLC purified, at prices ~50% of competitors´! Choosing transfection methods appropriate for each type of RNAi reagent is critical for successful gene silencing. The AvantGene Transfection reagent works well for delivering PCR or plasmid DNA reagents, whereas AvantGeneSi is ideal for synthetic siRNA. When primary cells need to be used, the choices for RNAi reagent delivery are normally narrowed down to electroporation or viral vectors, with the latter more widely used. Allele´s RNAi product family includes U6 promoter driven shRNA cassettes (US patents issued and pending) carried on HIV-based lentiviral vector pLICO (implico, Latin for intertwine), and MMLV-based retroviral vector pCHAC (Chac, Mayan for God of culture and growth), with the options of including an IRES-driven mWasabi.
